Demonstration of a high yield fermentation process for 2G bioethanol production

Using an optimal combination of recent danish developments in efficient technologies for pretreatment,
enzymatic hydrolysis and of modified fermenting yeast, 2. generation bioethanol can in theory at present
be produced in high yield from wheat straw at a competitive price. The project aim is to develop,
implement and demonstrate such a combination process in Inbicon's Kalundborg facility.

The aim of the project was to develop and demonstrate Terranols GM yeast for fermentation (fermentation) of C5 sugars in the 2nd generation bioethanol production in co-operation with DONG / Inbicon. In the project the yeast's performance has been successfully demonstrated in industrial scale integrated with the car's overall process for the production of 2nd generation bioethanol primarily with straw as feedstock.
Project description

The economic feasibility of a 2. generation bioethanol production process integrating the Inbicon pretreatment technology with highly robust Terranol yeast strains that, in addition to hexoses, also ferment xylose into ethanol with high yield and efficiency will be demonstrated.

Development of batch and fed-batch fermentation processes in hydrolysate slurry, followed by further development into an expected process in semi-continuous or continuous mode, and determination of process critical parameters will be performed. Also, a process monitoring system and a procedure for initial production of yeast inoculum will be implemented in large scale.

A number of specific modifications of the Inbicon facility are needed and will be addressed before the demonstration of the high yield fermentation technology can take place.

As the use of a Genetically Modified Organism (GMO) is required for the completion of this project, regulatory work necessary for obtaining permission to production with af GMO will also be addressed. This involves the complete description of the joint technology to be employed at the Inbicon facility.

Thus, the parameters for continued operation in industrial scale will be established and subsequently transferred into large scale thereby achieving at least 50% increased yield on wheat straw in the Kalundborg demonstration facility.

Results

Terranol and DONG Energy have aimed to develop, implement and test a combina-tion of 2G bioethanol production technologies for efficient pre-treatment, enzymatic hydrolysis and yeast fermentation. Proprietary technologies developed by the two companies have been joined in a successful demonstration of yeast based C6 and C5 sugar co-fermentation at DONG Energy's demonstration plant for Inbicon 2G ethanol technology in Kalundborg. The results in 270.000 litre scale are in accord-ance with results obtained in 2 litre laboratory scale. The Terranol yeast yields close to the theoretically maximum and is fast and robust in the Inbicon process. More than 90% of both C5 and C6 sugars was converted into ethanol within 48 hours using a low yeast pitch. Furthermore, a semi-continuous fermentation protocol de-veloped by Terranol with partial draining and refilling was demonstrated in the Inbi-con GMO approved pilot plant in Skærbæk in 300L scale. A very satisfactory ethanol yield of 93% was achieved
